SB 203580, an inhibitor of p38 MAPK, in the range 0.1\u20131 \u03bcM inhibited IL\u20101\u2010stimulated PGE<jats:sub>2<\/jats:sub> (but not arachidonic acid) release and this was associated with inhibition of induction of COX\u20102 protein and mRNA. IL\u20101 stimulated COX\u20102 transcription in HeLa cells about 2\u2010fold as judged by both reporter gene and nuclear run\u2010on assays. The inhibitor had no significant effect on this. However, in cells previously stimulated with IL\u20101 it caused rapid destabilisation of COX\u20102 mRNA independently of on\u2010going transcription.
