The t\u2010SNARE Sed5p is complexed with Sly1p in vivo. The mutant form Sly1\u201020p rescues Ypt1p\u2010lacking cells from lethality, suggesting an inhibitory function of Sly1p in v\u2010SNARE\/t\u2010SNARE interaction. Using surface plasmon resonance spectroscopy, we found that Sed5p binds Sly1p and Sly1\u201020p with equally high affinity (<jats:italic>K<\/jats:italic><jats:sub>D<\/jats:sub>=5.13\u00d710<jats:sup>\u22129<\/jats:sup>M and 4.74\u00d710<jats:sup>\u22129<\/jats:sup>M, respectively). Deletion studies show that the N\u2010terminal half of Sly1p rather than the C\u2010terminus (harbouring the E532K substitution in Sly1\u201020p) is most critical for its binding to Sed5p.
