Helminthol."],"published-print":{"date-parts":[[2016,1]]},"abstract":"<jats:title>Abstract<\/jats:title><jats:p><jats:italic>Meloidogyne hispanica<\/jats:italic> (<jats:italic>Mhi<\/jats:italic>) is a difficult-to-control polyphagous root-knot nematode (RKN) species of emerging importance for economically valuable crops. Nematode secretions are likely to be the first signals perceived by the plant and are thought to be involved in various aspects of the plant\u2013nematode interaction. The aims of this work were to identify and characterize <jats:italic>M. hispanica<\/jats:italic> parasitism genes: cathepsin L cysteine protease (<jats:italic>cpl-1<\/jats:italic>), calreticulin (<jats:italic>crt-1<\/jats:italic>), \u03b2-1,4-endoglucanase-1 (<jats:italic>eng-1<\/jats:italic>) and manganese superoxide dismutase (<jats:italic>mnsod<\/jats:italic>). As there are no genomic data available for <jats:italic>M. hispanica,<\/jats:italic> primers were designed from the conserved regions of the putative parasitism genes in <jats:italic>M.<\/jats:italic><jats:italic>incognita<\/jats:italic> and <jats:italic>M. hapla<\/jats:italic> and used to amplify the genes in <jats:italic>M. hispanica,<\/jats:italic> which led to the successful amplification of these genes in <jats:italic>M. hispanica.<\/jats:italic> Partial gene sequences were also obtained for <jats:italic>M. arenaria<\/jats:italic>, <jats:italic>M. hapla<\/jats:italic>, <jats:italic>M. hispanica<\/jats:italic>, <jats:italic>M. incognita<\/jats:italic> and <jats:italic>M. javanica<\/jats:italic><jats:italic>cpl-1<\/jats:italic>, <jats:italic>crt-1<\/jats:italic>, <jats:italic>eng-1<\/jats:italic> and <jats:italic>mnsod<\/jats:italic> genes, and their phylogenetic relationship analysed. In order to determine whether these genes are differentially expressed during <jats:italic>M. hispanica<\/jats:italic> development, cDNA was amplified from mRNA isolated from eggs, second-stage juveniles (J2) and females. Amplification products were observed from cDNA of all developmental stages for the <jats:italic>Mhi-cpl-1<\/jats:italic> and <jats:italic>Mhi-crt-1<\/jats:italic> genes. However, the gene <jats:italic>Mhi-crt-1<\/jats:italic> exhibited intense amplification bands in females, while the <jats:italic>Mhi-eng-1<\/jats:italic> gene was equally amplified in eggs and J2 and the <jats:italic>Mhi-mnsod<\/jats:italic> gene was only expressed in eggs.
