Genotypic relationships within the complex were evaluated at different \nlevels by: binding of the probe Lmet9, specific for <jats:italic>L. chagasi<\/jats:italic> and Old World <jats:italic>Leishmania<\/jats:italic> spp.; partial sequencing of a \nconstitutive major surface protease single gene (<jats:italic>mspC<\/jats:italic>) and random amplification of polymorphic DNA (RAPD). The Old \nWorld <jats:italic>Leishmania<\/jats:italic> spp. and the <jats:italic>L. donovani<\/jats:italic> complex have a monophyletic origin. <jats:italic>Leishmania chagasi<\/jats:italic> clearly belongs to \nthe <jats:italic>L. donovani<\/jats:italic> complex but it is indistinguishable from <jats:italic>L. infantum<\/jats:italic>, which suggests introduction of <jats:italic>L. chagasi<\/jats:italic> into the \nNew World in recent history. <jats:italic>Leishmania infantum<\/jats:italic>\/<jats:italic>L. chagasi<\/jats:italic> was identified as a monophyletic group within the <jats:italic>L. \ndonovani<\/jats:italic> complex but <jats:italic>L. donovani<\/jats:italic> may be paraphyletic.
