We have now pharmacologically characterized this receptor in rabbit mesenteric arteries and investigated how this adenosine receptor response changes in portal hypertensive animals since the adenosine response is decreased.<\/jats:p><\/jats:list-item>\n<jats:list-item><jats:p>The closest non\u2010metabolisable adenosine analogue, 2\u2010chloroadenosine (CADO), the mixed A<jats:sub>1<\/jats:sub>\/A<jats:sub>2<\/jats:sub> receptor agonist, 5\u2032\u2010ethylcarboxamidoadenosine (NECA), and the selective A<jats:sub>2A<\/jats:sub> receptor agonist, 2\u2010[4\u2010(2\u2010p\u2010carbonyethyl)phenylamino]\u20105\u2032\u2010N\u2010ethylcarboxamidoadenosine (CGS 21680) (1\u2003p<jats:sc>M<\/jats:sc>\u2003\u2013\u20031\u2003m<jats:sc>M<\/jats:sc>) relaxed noradrenaline pre\u2010contracted arteries with a rank order of potency of CGS 21680 (EC<jats:sub>50<\/jats:sub>=20\u2003n<jats:sc>M<\/jats:sc>)NECA (60\u2003n<jats:sc>M<\/jats:sc>)&gt;&gt;CADO (640\u2003n<jats:sc>M<\/jats:sc>).<\/jats:p><\/jats:list-item>\n<jats:list-item><jats:p>The selective A<jats:sub>2A<\/jats:sub> receptor antagonist, 4\u2010(2\u2010[7\u2010amino\u20102\u2010(2\u2010furyl)\u2010[1,2,4]\u2010triazolo[2,3\u2010a][1,3,5]\u2010triazin\u20105\u2010ylamino]ethyl)phenol (ZM 241385, 100\u2003n<jats:sc>M<\/jats:sc>), shifted to the right the CADO concentration\u2010response curve.<\/jats:p><\/jats:list-item>\n<jats:list-item><jats:p>In portal hypertensive animals, there was mainly a decreased potency but also a decreased efficacy of all tested adenosine agonists compared to normal animals.
