We investigated how pre\u2010 and postsynaptic modulation of GABAergic transmission (to both pyramidal cells and interneurones) by VIP could influence excitatory synaptic transmission in the CA1 area of the hippocampus.<\/jats:p><\/jats:list-item>\n<jats:list-item><jats:p>VIP (0.1\u2013100 n<jats:sc>M<\/jats:sc>) increased [<jats:sup>3<\/jats:sup>H]GABA release from hippocampal synaptosomes (maximum effect at 1 n<jats:sc>M<\/jats:sc> VIP; 63.8\u00b14.0%) but did not change [<jats:sup>3<\/jats:sup>H]glutamate release.<\/jats:p><\/jats:list-item>\n<jats:list-item><jats:p>VIP (0.3\u201330 n<jats:sc>M<\/jats:sc>) enhanced synaptic transmission in hippocampal slices (maximum effect at 1 n<jats:sc>M<\/jats:sc> VIP; field excitatory postsynaptic potentials (epsp) slope: 23.7\u00b11.1%; population spike amplitude: 20.3\u00b11.7%). The action on field epsp slope was fully dependent on GABAergic transmission since it was absent in the presence of picrotoxin (50 <jats:italic>\u03bc<\/jats:italic><jats:sc>M<\/jats:sc>) plus CGP55845 (1 <jats:italic>\u03bc<\/jats:italic><jats:sc>M<\/jats:sc>).<\/jats:p><\/jats:list-item>\n<jats:list-item><jats:p>VIP (1 n<jats:sc>M<\/jats:sc>) did not change paired\u2010pulse facilitation but increased paired\u2010pulse inhibition in CA1 pyramidal cells (16.0\u00b10.9%), reinforcing the involvement of GABAergic transmission in the action of VIP.<\/jats:p><\/jats:list-item>\n<jats:list-item><jats:p>VIP (1 n<jats:sc>M<\/jats:sc>) increased muscimol\u2010evoked inhibitory currents by 36.4\u00b18.7% in eight out of ten CA1 interneurones in the <jats:italic>stratum radiatum<\/jats:italic>.
