{"status":"ok","message-type":"work","message-version":"1.0.0","message":{"indexed":{"date-parts":[[2026,3,18]],"date-time":"2026-03-18T03:04:17Z","timestamp":1773803057044,"version":"3.50.1"},"reference-count":0,"publisher":"Portland Press Ltd.","issue":"1","content-domain":{"domain":["portlandpress.com"],"crossmark-restriction":true},"short-container-title":[],"published-print":{"date-parts":[[1998,5,15]]},"abstract":"<jats:p>We have used two hydroxylated naphthoquinol menaquinol analogues, reduced plumbagin (PBH2, 5-hydroxy-2-methyl-1,4-naphthoquinol) and reduced lapachol [LPCH2, 2-hydroxy-3-(3-methyl-2-butenyl)-1,4-naphthoquinol], as substrates for Escherichia colianaerobic reductases. These compounds have optical, solubility and redox properties that make them suitable for use in studies of the enzymology of menaquinol oxidation. Oxidized plumbagin and oxidized lapachol have well resolved absorbances at 419\u00a0nm (\u03b5 = 3.95\u00a0mM-1\u00b7cm-1) and 481\u00a0nm (\u03b5 = 2.66\u00a0mM-1\u00b7cm-1) respectively (in Mops\/KOH buffer, pH\u00a07.0). PBH2 is a good substrate for nitrate reductase A (Km = 282\u00b128\u00a0\u00b5M, kcat = 120\u00b16\u00a0s-1) and fumarate reductase (Km = 155\u00b124\u00a0\u00b5M, kcat = 30\u00b12\u00a0s-1), but not for DMSO reductase. LPCH2 is a good substrate for nitrate reductase A (Km = 57\u00b135\u00a0\u00b5M, kcat = 68\u00b113\u00a0s-1), fumarate reductase (Km = 85\u00b127\u00a0\u00b5M, kcat = 74\u00b16\u00a0s-1) and DMSO reductase (Km = 238\u00b130\u00a0\u00b5M, kcat = 191\u00b121\u00a0s-1).
