To elucidate the role of several\n key proteins involved in this process, we have analysed the time dependence of replication\n restart in strains carrying a combination of mutations in <jats:italic>lexA<\/jats:italic>, <jats:italic>recA<\/jats:italic>,\n <jats:italic>polB<\/jats:italic> (pol II), <jats:italic>umuDC<\/jats:italic> (pol V), <jats:italic>priA<\/jats:italic>, <jats:italic>dnaC<\/jats:italic>, <jats:italic>recF<\/jats:italic>,\n <jats:italic>recO<\/jats:italic> or <jats:italic>recR<\/jats:italic>. We find that both pol II and the origin\u2010independent primosome\u2010assembling\n function of PriA are essential for the immediate recovery of DNA synthesis after\n UV irradiation. In their absence, translesion replication or \u2018replication\n readthrough\u2019 occurs <jats:bold>\u2248<\/jats:bold> 50 min after UV and is pol V\u2010dependent. In\n a wild\u2010type, <jats:italic>lexA<\/jats:italic><jats:bold><jats:sup>+<\/jats:sup><\/jats:bold> background, mutations in <jats:italic>recF<\/jats:italic>,\n <jats:italic>recO<\/jats:italic> or <jats:italic>recR<\/jats:italic> block both pathways. Similar results were obtained with\n a <jats:italic>lexA<\/jats:italic>(Def) <jats:italic>recF<\/jats:italic> strain. However, <jats:italic>lexA<\/jats:italic>(Def) <jats:italic>recO<\/jats:italic> or <jats:italic>lexA<\/jats:italic>(Def)\n <jats:italic>recR<\/jats:italic> strains, although unable to facilitate PriA\u2010pol II\u2010dependent restart,\n were able to perform pol V\u2010dependent readthrough. The defects in restart attributed\n to mutations in <jats:italic>recF<\/jats:italic>, <jats:italic>recO<\/jats:italic> or <jats:italic>recR<\/jats:italic> were suppressed in a <jats:italic>recA730\n lexA<\/jats:italic>(Def) strain expressing constitutively activated RecA (RecA*). Our data suggest\n that in a wild\u2010type background, RecF, O and R are important for the induction of\n the SOS response and the formation of RecA*\u2010dependent recombination intermediates\n necessary for PriA\/Pol II\u2010dependent replication restart.
