TNF\u03b1 may protect neurons against excitotoxic and oxidative insults by a mechanism involving activation of the transcription factor NF\u2010\u03baB. Whole\u2010cell perforated patch clamp recordings in cultured rat hippocampal neurons showed that long\u2010term treatment (24\u201348 h) with TNF\u03b1 increases Ca<jats:sup>2+<\/jats:sup> current density; pharmacological analysis indicated a major increase in current through L\u2010type voltage\u2010dependent calcium channels. Long\u2010term treatment with TNF\u03b1 caused a decrease in currents induced by glutamate, NMDA, AMPA, and kainate. Shorter exposures to TNF\u03b1 (acute; 2 h) did not alter Ca<jats:sup>2+<\/jats:sup> current or glutamate receptor agonist\u2010induced currents. Ceramide, an intracellular messenger that activates the transcription factor NF\u2010\u03baB, mimicked the actions of TNFs on Ca<jats:sup>2+<\/jats:sup> current density and currents induced by glutamate receptor agonists. Cotreatment with \u03baB decoy DNA abolished the effects of TNF\u03b1 on Ca<jats:sup>2+<\/jats:sup> current and excitatory amino acid\u2010induced currents, demonstrating a requirement for NF\u2010\u03baB activation in the actions of TNF\u03b1. Neurons pretreated with TNF\u03b1 exhibited increased intracellular Ca<jats:sup>2+<\/jats:sup> concentrations following membrane depolarization but reduced intracellular Ca<jats:sup>2+<\/jats:sup> concentration responses to excitatory amino acids, compared with neurons in untreated control cultures or cultures cotreated with \u03baB decoy DNA. These findings suggest important roles for the transcription factor NF\u2010\u03baB in modulation of voltage\u2010dependent calcium channels and glutamate receptors and the many physiological and pathophysiological processes in which these ion channels are involved.
