Although their biosynthesis by means of mevalonate has been studied in detail, a second biosynthetic pathway for their formation by means of 1-deoxy-\n            <jats:sc>d<\/jats:sc>\n            -xylulose 5-phosphate has been discovered only recently in plants and certain eubacteria. Earlier\n            <jats:italic>in vivo<\/jats:italic>\n            experiments with recombinant\n            <jats:italic>Escherichia coli<\/jats:italic>\n            strains showed that exogenous 1-deoxy-\n            <jats:sc>d<\/jats:sc>\n            -xylulose can be converted into 1-hydroxy-2-methyl-2-(\n            <jats:italic>E<\/jats:italic>\n            )-butenyl 4-diphosphate by the consecutive action of enzymes specified by the\n            <jats:italic>xylB<\/jats:italic>\n            and\n            <jats:italic>ispCDEFG<\/jats:italic>\n            genes. This article describes the transformation of exogenous [U-\n            <jats:sup>13<\/jats:sup>\n            C\n            <jats:sub>5<\/jats:sub>\n            ]1-deoxy-\n            <jats:sc>d<\/jats:sc>\n            -xylulose into a 5:1 mixture of [U-\n            <jats:sup>13<\/jats:sup>\n            C\n            <jats:sub>5<\/jats:sub>\n            ]isopentenyl diphosphate and [U-\n            <jats:sup>13<\/jats:sup>\n            C\n            <jats:sub>5<\/jats:sub>\n            ]dimethylallyl diphosphate by an\n            <jats:italic>E. coli<\/jats:italic>\n            strain engineered for the expression of the\n            <jats:italic>ispH (lytB)<\/jats:italic>\n            gene in addition to recombinant\n            <jats:italic>xylB<\/jats:italic>\n
