U.S.A."],"published-print":{"date-parts":[[2000,12,19]]},"abstract":"<jats:p>\n                    Meiosis is the developmental program by which diploid\n organisms produce haploid gametes capable of sexual reproduction. Here\n we describe the yeast gene\n                    <jats:italic>AMA1<\/jats:italic>\n                    , a new member of the\n Cdc20 protein family that regulates the multisubunit ubiquitin ligase\n termed the anaphase promoting complex\/cyclosome (APC\/C).\n                    <jats:italic>AMA1<\/jats:italic>\n                    is developmentally regulated in that its\n transcription and splicing occur only in meiotic cells. The\n meiosis-specific processing of\n                    <jats:italic>AMA1<\/jats:italic>\n                    mRNA depends on the\n previously described\n                    <jats:italic>MER1<\/jats:italic>\n                    splicing factor. Several\n results indicate that Ama1p is required for APC\/C function during\n meiosis. First, coimmunoprecipitation assays indicate that Ama1p\n associates with the APC\/C\n                    <jats:italic>in vivo<\/jats:italic>\n                    . Second, Ama1p is\n required for the degradation of the B-type cyclin Clb1p, an APC\/C\n substrate in both meiotic and mitotic cells. Third, ectopic\n overexpression of\n                    <jats:italic>AMA1<\/jats:italic>\n                    is able to stimulate\n ubiquitination of Clb1p\n                    <jats:italic>in vitro<\/jats:italic>\n                    and degradation of\n Clb1p\n                    <jats:italic>in vivo<\/jats:italic>\n                    . Mutants lacking\n                    <jats:italic>AMA1<\/jats:italic>\n                    revealed that it is required for the first meiotic division but not the\n mitotic-like meiosis II. In addition,\n                    <jats:italic>ama1<\/jats:italic>\n                    mutants are\n defective for both spore wall assembly and the expression of late\n meiotic genes.
