U.S.A."],"published-print":{"date-parts":[[2000,12,5]]},"abstract":"<jats:p>\n            The\n            <jats:italic>Saccharomyces cerevisiae<\/jats:italic>\n            proteins Tel1p and\n Mec1p are involved in telomere length regulation and cellular responses\n to DNA damage. The closest relative of these proteins is the human\n            <jats:italic>A<\/jats:italic>\n            taxia\n            <jats:italic>T<\/jats:italic>\n            elangiectasia\n            <jats:italic>M<\/jats:italic>\n            utated (ATM) protein, a wortmannin-sensitive protein\n kinase that primarily phosphorylates serines in an SQ motif. We\n constructed yeast strains containing functional epitope-tagged versions\n of Tel1p and Mec1p. We showed that immunoprecipitated Tel1p and Mec1p\n were capable of\n            <jats:italic>in vitro<\/jats:italic>\n            phosphorylation of the\n mammalian protein PHAS-I (\n            <jats:italic>P<\/jats:italic>\n            hosphorylated\n            <jats:italic>H<\/jats:italic>\n            eat and\n            <jats:italic>A<\/jats:italic>\n            cid\n            <jats:italic>S<\/jats:italic>\n            table\n protein). These activities are sensitive to wortmannin. Tel1p\n phosphorylates serine in an SQ motif in PHAS-I. Mutations in the kinase\n domains of Tel1p and Mec1p result in loss of\n            <jats:italic>in vitro<\/jats:italic>\n            kinase activity and the\n            <jats:italic>in vivo<\/jats:italic>\n            phenotypes associated\n with the null\n            <jats:italic>tel1<\/jats:italic>\n            and\n            <jats:italic>mec1<\/jats:italic>\n            mutations.\n
