In this work, we investigated the effect of dexamethasone on NO production induced by the epidermal cytokine granulocyte\u2010macrophage colony\u2010stimulating factor (GM\u2010CSF) in a mouse fetal skin dendritic cell line.<\/jats:p><jats:p><jats:italic>Methods:<\/jats:italic> NO production was assessed by the method of Griess. Expression of the inducible isoform of nitric oxide synthase (iNOS) protein was evaluated by western blot analysis and immunofluorescence microscopy. Western blot analysis was also performed to evaluate cytosolic IkappaB\u2010alpha (I\u03baB\u2010\u03b1) protein levels. The electrophoretic mobility shift assay was used to evaluate the activation or inhibition of nuclear factor kappa B (NF\u2010\u03baB).<\/jats:p><jats:p><jats:italic>Results:<\/jats:italic> GM\u2010CSF induced iNOS expression and NO production, and activated the transcription factor NF\u2010\u03baB. Dexamethasone inhibited, in a dose\u2010dependent manner, NO production induced by GM\u2010CSF. Addition of dexamethasone to the culture, 30 min before GM\u2010CSF stimulation, significantly inhibited the cellular expression of iNOS. Dexamethasone also inhibited GM\u2010CSF\u2010induced NF\u2010\u03baB activation by preventing a significant decrease on the I\u03baB\u2010\u03b1 protein levels, thus blocking NF\u2010\u03baB migration to the nucleus.<\/jats:p><jats:p><jats:italic>Conclusions:<\/jats:italic> The corticosteroid dexamethasone inhibits GM\u2010CSF\u2010induced NF\u2010\u03baB activation, iNOS protein expression and NO production.
