The Qa-1b peptide-binding site is predominantly occupied by a single nonameric peptide, Qa-1 determinant modifier (Qdm), derived from the leader sequence of H-2D and L molecules. Five anchor residues were identified in this study by measuring the peptide-binding affinities of substituted Qdm peptides in experiments with purified recombinant Qa-1b. A candidate peptide-binding motif was determined by sequence analysis of peptides eluted from Qa-1 that had been folded in the presence of random peptide libraries or pools of Qdm derivatives randomized at specific anchor positions. The results indicate that Qa-1b can bind a diverse repertoire of peptides but that Qdm has an optimal primary structure for binding Qa-1b. Flow cytometry experiments with Qa-1b tetramers and NK target cell lysis assays demonstrated that CD94\/NKG2A discriminates between Qa-1b complexes containing peptides with substitutions at nonanchor positions P4, P5, or P8. Our findings suggest that it may be difficult for viruses to generate decoy peptides that mimic Qdm and raise the possibility that competitive replacement of Qdm with other peptides may provide a novel mechanism for activation of NK cells.<\/jats:p>","DOI":"10.1084\/jem.192.5.613","type":"journal-article","created":{"date-parts":[[2002,7,26]],"date-time":"2002-07-26T16:48:33Z","timestamp":1027702113000},"page":"613-624","update-policy":"https:\/\/doi.org\/10.1084\/jem.crossmarkpolicy","source":"Crossref","is-referenced-by-count":80,"title":["Analysis of Qa-1bPeptide Binding Specificity and the Capacity of Cd94\/Nkg2a to Discriminate between Qa-1\u2013Peptide Complexes"],"prefix":"10.1084","volume":"192","author":[{"given":"Jennifer R.","family":"Kraft","sequence":"first","affiliation":[{"name":"aDepartment of Pathology and Laboratory Medicine, Emory University School of Medicine, Atlanta, Georgia 30322"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"given":"Russell E.","family":"Vance","sequence":"additional","affiliation":[{"name":"cDepartment of Molecular and Cell Biology and Cancer Research Laboratory, University of California at Berkeley, Berkeley, California 94720"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"given":"Jan","family":"Pohl","sequence":"additional","affiliation":[{"name":"bMicrochemical Facility, Emory University School of Medicine, Atlanta, Georgia 30322"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"given":"Amy M.","family":"Martin","sequence":"additional","affiliation":[{"name":"bMicrochemical Facility, Emory University School of Medicine, Atlanta, Georgia 30322"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"given":"David H.","family":"Raulet","sequence":"additional","affiliation":[{"name":"cDepartment of Molecular and Cell Biology and Cancer Research Laboratory, University of California at Berkeley, Berkeley, California 94720"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"given":"Peter E.","family":"Jensen","sequence":"additional","affiliation":[{"name":"aDepartment of Pathology and Laboratory Medicine, Emory University School of Medicine, Atlanta, Georgia 30322"}],"role":[{"role":"author","vocabulary":"crossref"}]}],"member":"291","published-online":{"date-parts":[[2000,8,28]]},"reference":[{"key":"2023072509241373900_R1","doi-asserted-by":"crossref","first-page":"525","DOI":"10.1007\/BF01576982","article-title":"A new serologically defined locus, Qa-1, in the Tla-region of the mouse","volume":"3","author":"Stanton","year":"1976","journal-title":"Immunogenetics"},{"key":"2023072509241373900_R2","doi-asserted-by":"crossref","first-page":"469","DOI":"10.1016\/S0092-8674(85)80020-9","article-title":"Expression of class I genes in the major histocompatibility complexidentification of eight distinct mRNAs in DBA\/2 mouse liver","volume":"41","author":"Lalanne","year":"1985","journal-title":"Cell"},{"key":"2023072509241373900_R3","doi-asserted-by":"crossref","first-page":"341","DOI":"10.1084\/jem.166.2.341","article-title":"A low polymorphic mouse H-2 class I gene from the Tla complex is expressed in a broad variety of cell types","volume":"166","author":"Transy","year":"1987","journal-title":"J.
