The resistance mechanism is unknown, but an efficient reactive oxygen species (ROS) scavenging system and DNA-repair and DNA-protection mechanisms are believed to play important roles. Here, the cloning and small- and medium-scale expression tests of a novel dye-decolourizing peroxidase from <jats:italic>D. radiodurans<\/jats:italic> (<jats:italic>Dr<\/jats:italic>DyP) using three different <jats:italic>Escherichia coli<\/jats:italic> strains and three different temperatures in order to identify the optimum conditions for the expression of recombinant <jats:italic>Dr<\/jats:italic>DyP are presented. The best expression conditions were used for large-scale expression and yielded \u223c10\u2005mg recombinant <jats:italic>Dr<\/jats:italic>DyP per litre of culture after purification. Initial characterization experiments demonstrated unusual features with regard to the haem spin state, which motivated the crystallization experiment. The obtained crystals were used for data collection and diffracted to 2.2\u2005\u00c5 resolution. The crystals belonged to the trigonal space group <jats:italic>P<\/jats:italic>3<jats:sub>1<\/jats:sub> or <jats:italic>P<\/jats:italic>3<jats:sub>2<\/jats:sub>, with unit-cell parameters <jats:italic>a<\/jats:italic>\u00a0=\u00a0<jats:italic>b<\/jats:italic>\u00a0=\u00a064.13, <jats:italic>c<\/jats:italic>\u00a0=\u00a0111.32\u2005\u00c5, and are predicted to contain one <jats:italic>Dr<\/jats:italic>DyP molecule per asymmetric unit.
