{"status":"ok","message-type":"work","message-version":"1.0.0","message":{"indexed":{"date-parts":[[2025,12,14]],"date-time":"2025-12-14T12:03:24Z","timestamp":1765713804859},"reference-count":28,"publisher":"Wiley","issue":"4","license":[{"start":{"date-parts":[[2005,3,23]],"date-time":"2005-03-23T00:00:00Z","timestamp":1111536000000},"content-version":"vor","delay-in-days":0,"URL":"http:\/\/onlinelibrary.wiley.com\/termsAndConditions#vor"}],"content-domain":{"domain":["onlinelibrary.wiley.com"],"crossmark-restriction":true},"short-container-title":["Histopathology"],"published-print":{"date-parts":[[2005,4]]},"abstract":"<jats:p><jats:bold>Aims<\/jats:bold> :\u2002To evaluate the HER\u20102\/<jats:italic>neu<\/jats:italic> status at the mRNA and DNA level of breast carcinomas and to compare it with HER\u20102\/<jats:italic>neu<\/jats:italic> receptor overexpression by immunohistochemistry (IHC).<\/jats:p><jats:p><jats:bold>Methods and results<\/jats:bold> :\u2002In 32 invasive breast carcinomas, frozen tissue was available for real\u2010time detection of HER\u20102\/<jats:italic>neu<\/jats:italic> mRNA levels by reverse transcription\u2010polymerase chain reaction (RT\u2010PCR). Corresponding paraffin sections were examined by IHC and fluorescence <jats:italic>in\u2010situ<\/jats:italic> hybridization (FISH). Thereby, different IHC and FISH procedures were compared. Using microwave epitope retrieval, all 32 cases scored 3+ on IHC, whereas only 28 out of 32 cases scored IHC 3+ using water bath epitope retrieval. All of these 28 cases showed increased levels of HER\u20102\/<jats:italic>neu<\/jats:italic> mRNA. Dual\u2010colour FISH analysis showed corresponding gene amplification in all 28 cases, with two cases showing a peculiar amplification pattern. In the remaining four cases, scoring IHC 2+ using water bath epitope retrieval, mRNA levels were not elevated. Three cases did not have gene amplification and one case showed low\u2010level HER\u20102\/<jats:italic>neu<\/jats:italic> gene amplification. All four carcinomas showed chromosome 17 polysomy.<\/jats:p><jats:p><jats:bold>Conclusions<\/jats:bold> :\u2002Real\u2010time RT\u2010PCR is accurate in selecting breast carcinoma cases scoring 3+ by IHC with high\u2010level gene amplification.
