The tetracycline efflux proteins fall into three families: (i) those encoded by pBR322, RP1, and Tn 10 (<jats:italic>Escherichia coli<\/jats:italic>); (ii) pT181 (<jats:italic>Staphyloccoccus aureus<\/jats:italic>) and pTHT15 (<jats:italic>Bacillus subtilis<\/jats:italic>); and (iii) <jats:italic>tet<\/jats:italic>347 (Streptomyces rimosus). There is global sequence homology within each of the first two families, but there is none between the families. The pT181\/pTHT15 family shares close homology with the <jats:italic>N<\/jats:italic>\u2010terminal half of the methlenomycin A efflux protein (<jats:italic>Streptomyces coelicor<\/jats:italic>), while <jats:italic>tet<\/jats:italic>347 ressembles the <jats:italic>C<\/jats:italic>\u2010terminal half. Portions of the <jats:italic>N<\/jats:italic>\u2010terminal half of the <jats:italic>Tn<\/jats:italic>10\u2010encoded protein show significant resemblance to portions in the <jats:italic>N<\/jats:italic>\u2010terminal half of the pT181\/pTHT15 family, but this sometimes occurs among transport proteins which do not have a common subtrate.
