In this report, we have examined the mechanism of resistance to streptomycin (Sm) in <jats:italic>M. tuberculosis<\/jats:italic> through the cloning and nucleotide sequence analysis of the gene encoding the ribosomal S<jats:sup>R<\/jats:sup> protein (<jats:italic>rpsL<\/jats:italic> gene) from streptomycin\u2010resistant strains and their streptomycin\u2010sensitive parental strains. We have demonstrated that five singly Sm<jats:sup>R<\/jats:sup><jats:italic>M. tuberculosis<\/jats:italic> strains and an Sm<jats:sup>R<\/jats:sup> isolate that has reduced sensitivity to multiple antibiotics have identical point mutations at codon 43 of the <jats:italic>rpsL<\/jats:italic> gene.
