(1992) <jats:italic>J. Biol. Chem. 267<\/jats:italic>, 25927\u201325933]. The Ca<jats:sup>2+<\/jats:sup> sensitivity of the lung \u03b1\u2010actinin for the interaction with polymerized actin (F\u2010actin) was much lower than those of the other reported non\u2010muscle \u03b1\u2010actinins. Here, we isolated a cDNA clone encoding the novel \u03b1\u2010actinin by screening a chicken lung \u03b3gt11 cDNA library with antibody specific for the low\u2010Ca<jats:sup>2+<\/jats:sup>\u2010sensitive \u03b1\u2010actinin. The deduced amino acid sequence of the lung \u03b1\u2010actinin showed 76%, 82% and 83% identity to those of chicken skeletal\u2010muscle, smooth\u2010muscle and fibroblast\u2010type \u03b1\u2010actinin, respectively. Marked difference in the structure between the lung\u2010type and the other \u03b1\u2010actinins was found in the extreme NH<jats:sub>2<\/jats:sub>\u2010terminal and in the COOH\u2010terminal half; in the third and fourth regions of four spectrin\u2010like repeats, and in two Ca<jats:sup>2+<\/jats:sup>\u2010binding EF\u2010hand consensus regions. The NH<jats:sub>2<\/jats:sub>\u2010terminal\u2010side EF\u2010hand contained a notable defect in one of the five oxygen\u2010containing amino acid side chains involved in chelating Ca<jats:sup>2+<\/jats:sup>, suggesting that the lower Ca<jats:sup>2+<\/jats:sup> sensitivity of the lung \u03b1\u2010actinin is ascribable to this defect. Northern blot analysis showed that the expression pattern of lung\u2010type \u03b1\u2010actinin mRNA in various non\u2010muscle tissues differed from that of the other known non\u2010muscle\u2010type (fibroblast\u2010type) \u03b1\u2010actinin.
