They were functionally solubilized by the non\u2010denaturing zwitterionic detergent 3\u2010[(3\u2010cholamidopropyl)dimethylammonio]\u20101\u2010propanesulfonic acid (Chaps, 20 mM, detergent\/protein mass ratio 4:1) at a yield of 75\u201380%. Using [<jats:sup>3<\/jats:sup>H]progesterone as radioligand, binding studies showed high\u2010affinity and low\u2010affinity binding sites in microsomal preparations with an apparent <jats:italic>K<\/jats:italic><jats:sub>d1<\/jats:sub> of 11 nM and an apparent <jats:italic>K<\/jats:italic><jats:sub>d2<\/jats:sub> of 286 nM. In solubilized fractions the high\u2010affinity binding sites were present at an apparent <jats:italic>K<\/jats:italic><jats:sub>d<\/jats:sub> of 69 nM. In both preparations, progesterone binding was time\u2010dependent, saturable, reversible, and showed a similar hierachy of affinities for related steroids. A purification scheme was developed based on anion\u2010exchanger procedures. The purified fraction as identified by maximum specific progesterone\u2010binding activity contained two major polypeptides of apparent molecular masses (SDS\/PAGE) of 28 kDa and 56 kDa, respectively.
