Although levels of galactose and galactitol were markedly elevated, UDP\u2010galactose and UDP\u2010glucose levels were not significantly increased. The level of galactose 1\u2010phosphate rose to 1\u20103 \u03bcg\/g of fresh tissue by 14 h but gradually diminished until, at 48 h, the content was 0\u201025 \u03bcg\/g. The metabolic turnover of these compounds, as shown by labelling experiments with inorganic [<jats:sup>32<\/jats:sup>P]phosphate and [U\u2010<jats:sup>14<\/jats:sup>C]galactose, indicated that galactose 1\u2010phosphate and UDP\u2010galactose were rapidly metabolized, yet relatively little galactose was utilized by the brain as a source of energy. These observations have prompted us to propose a mechanism for the turnover of galactose 1\u2010phosphate that involves cyclical phosphorylation and dephosphorylation reactions in the brains of galactose\u2010fed chicks. In support of this hypothesis, we have identified phosphatase activity which has a relatively low <jats:italic>K<jats:sub>m<\/jats:sub><\/jats:italic> value for galactose 1\u2010phosphate (0\u201006\u20100\u201007 mM) in virtually all subcellular fractions of homogenates of chick brain.
