{"status":"ok","message-type":"work","message-version":"1.0.0","message":{"indexed":{"date-parts":[[2025,10,17]],"date-time":"2025-10-17T13:28:32Z","timestamp":1760707712189},"reference-count":43,"publisher":"Wiley","issue":"3","license":[{"start":{"date-parts":[[2006,10,4]],"date-time":"2006-10-04T00:00:00Z","timestamp":1159920000000},"content-version":"vor","delay-in-days":11721,"URL":"http:\/\/onlinelibrary.wiley.com\/termsAndConditions#vor"}],"content-domain":{"domain":[],"crossmark-restriction":false},"short-container-title":["Journal of Neurochemistry"],"published-print":{"date-parts":[[1974,9]]},"abstract":"<jats:p><jats:bold>Abstract\u2014<\/jats:bold> A new combined ion\u2010exchange and thin\u2010layer\u2010chromatographic procedure is described which separates and measures quantitatively, after intraventricular injection of [<jats:sup>3<\/jats:sup>H]dopamine (DA), the rat brain content of labelled noradrenaline (NA) and the following labelled noradrenaline metabolites: free 3\u2010methoxy\u20104\u2010hydroxyphenylethyleneglycol (MOPEG), conjugated MOPEG, free plus conjugated dihydroxyphenylethyleneglycol (DOPEG), vanillic mandelic acid (VMA) and normetanephrine (NM). Labelled dopamine and its metabolites were also measured. The time\u2010course study performed from 5 min to 24 h after [<jats:sup>3<\/jats:sup>H]DA showed that MOPEG and DOPEG, mainly as conjugates, are major NA metabolites whereas VMA is a very insignificant NA metabolite in the rat brain. A very rapid initial increase of [<jats:sup>3<\/jats:sup>H]NM, free MOPEG and conjugated MOPEG was found during the time interval where the [<jats:sup>3<\/jats:sup>H]NA biosynthesis is very high (0\u201315 min). This combined with the finding that these metabolites stabilize at lower levels during the [<jats:sup>3<\/jats:sup>H]NA \u2018storage phase\u2019 (9\u201324 h) provides a strong indication that newly synthesized NA preferentially is metabolized. Our measurements of endogenous NA, free MOPEG and conjugated MOPEG provide additional support. The injections of various decreasing doses of [<jats:sup>3<\/jats:sup>H]DA (3\u00b708\u20130\u00b70010 \u03bcg) showed that the proportions of total [<jats:sup>3<\/jats:sup>H]MOPEG and total [<jats:sup>3<\/jats:sup>H]DOPEG to [<jats:sup>3<\/jats:sup>H]NA were constant after all [<jats:sup>3<\/jats:sup>H]DA doses investigated. This finding indicates that the [<jats:sup>3<\/jats:sup>H]NA synthesized <jats:italic>in situ<\/jats:italic> behaves as a tracer, even after injections of non\u2010tracer doses of [<jats:sup>3<\/jats:sup>H]DA.
