Quantitation of both <jats:sup>14<\/jats:sup>CO<jats:sub>2<\/jats:sub> and [<jats:sup>14<\/jats:sup>C]GABA produced in radiometric assays from [U\u2010<jats:sup>14<\/jats:sup>CJglutamic acid has shown that measurement of <jats:sup>14<\/jats:sup>CO<jats:sub>2<\/jats:sub> evolution alone is not, in all cases, a valid estimate of true GAD activity. As evidenced by increased <jats:sup>,14<\/jats:sup>CO<jats:sub>2<\/jats:sub> production upon addition of NAD and CoA to assay mixtures, radiometric assay of GAD activity in crude homogenates may yield <jats:sup>14<\/jats:sup>CO<jats:sub>2<\/jats:sub> via the coupled reactions of glutamic acid dehydrogenase and a\u2010ketoglutarate dehydrogenase. The addition of 1 mM aminooxyacetic acid (AOAA) to assays of kidney homogenates inhibited [<jats:sup>,14<\/jats:sup>C]GABA production 92 per cent while <jats:sup>14<\/jats:sup>CO<jats:sub>2<\/jats:sub> production was inhibited only 53 per cent. No evidence was found to confirm the reported existence of a second form of the enzyme, GAD II. previously described by Haber <jats:italic>el al.<\/jats:italic> (H<jats:sc>aber<\/jats:sc> B., K<jats:sc>uriyama<\/jats:sc> K. &amp; R<jats:sc>oberts<\/jats:sc> E. (1970) <jats:italic>Biochem. Pharmac.<\/jats:italic> 19, 1119\u20101136).
