Using dialytrodes in the CA1 of the rat, we have investigated multiple\u2010unit activity and extracellular changes in acidic sulfur\u2010containing amino acids and\u2010\u03b3\u2010glutamyl peptides during ischemia (20\u2010min, four\u2010vessel occlusion) and during 8 h of reflow. Multiple\u2010unit activity was abolished during ischemia and for the following 1h, but then recovered, gradually reaching preischemic levels after 8 h of reflow. Extracellular cysteate, cysteine sulfinate, and \u03b3\u2010glutamyltaurine increased (1.5\u2010to threefold) during ischemia, and extracellular glutathione and \u03b3\u2010glutamylaspartate plus \u03b3\u2010glutamylglutamine increased during early reflow (two\u2010to threefold). The recovery of neuronal activity at 4\u20138 h was paralleled by an increase in extracellular cysteine sulfinate (2.5\u2010fold at 8 h of reflow). Perfusion with 10 \u03bc<jats:italic>M<\/jats:italic> tetrodotoxin at 8 h of reflow abolished the multiple\u2010unit activity and reduced extracellular cysteine sulfinate.
