The spontaneous efflux was biphasic, consisting of an initial rapid efflux followed by a much slower release.<\/jats:p><\/jats:list-item><jats:list-item><jats:p>The slices were depolarized by the addition to the medium of high potassium concentrations, ouabain or veratrine.<\/jats:p><\/jats:list-item><jats:list-item><jats:p>At concentrations greater than 30 mM, potassium evoked a striking increase in the release of [<jats:sup>3<\/jats:sup>H]\u2010spermine. Following uptake in the presence of 5.7 \u00d7 10<jats:sup>\u22129<\/jats:sup>M[<jats:sup>3<\/jats:sup>H]\u2010spermine, K<jats:sup>+<\/jats:sup>\u2010evoked release was dependent on the presence of calcium ions. Release of spermine after uptake at 5.6 \u00d7 10<jats:sup>\u22128<\/jats:sup>m or 5.0 \u00d7 10<jats:sup>\u22127<\/jats:sup>m was not calcium\u2010dependent.<\/jats:p><\/jats:list-item><jats:list-item><jats:p>The calcium\u2010dependent, K<jats:sup>+<\/jats:sup>\u2010stimulated release of spermine was inhibited in the presence of diphenylhydantoin (5 \u00d7 10<jats:sup>\u22123<\/jats:sup>m) or ruthenium red (10<jats:sup>\u22123<\/jats:sup>m).<\/jats:p><\/jats:list-item><jats:list-item><jats:p>Following uptake of 5.7 \u00d7 10<jats:sup>\u22129<\/jats:sup>m [<jats:sup>3<\/jats:sup>H]\u2010spermine in a sodium\u2010free medium, the calcium\u2010dependent, K<jats:sup>+<\/jats:sup>\u2010stimulated release was significantly inhibited.<\/jats:p><\/jats:list-item><jats:list-item><jats:p>Ouabain (10<jats:sup>\u22124<\/jats:sup>m) caused a large but calcium\u2010independent increase in the efflux of [<jats:sup>3<\/jats:sup>H]\u2010spermine.<\/jats:p><\/jats:list-item><jats:list-item><jats:p>Veratrine\u2010induced release was less substantial but was increased in a calcium\u2010free medium.
