Rat brain regions were examined for altered protein phosphotyrosines, using Western blot analysis and microwave irradiation to limit postmortem alterations, after administration of two convulsants: lithium plus pilocarpine or kainic acid (KA). Most phospho\u2010tyrosine proteins were unaltered by these treatments, but there was a large, specific increase in the tyrosine phosphorylation of a 40\u2010Kd protein. This increase was evident in all three regions examined: cerebral cortex, hippocampus, and striatum; it occurred abruptly with onset of generalized status epilepticus (SE) and remained elevated for at least 90 min. Most of the tyrosine phosphorylated 40\u2010Kd protein was in the cytosolic fraction. These results demonstrate a large, specific effect of chemically induced seizures on a single phosphotyrosine protein in rat brain.<\/jats:p><\/jats:sec><jats:sec><jats:title>R\u00c9SUM\u00c9<\/jats:title><jats:p>La phosphorylation des tyrosines prot\u00e9iques est un processus de modulation important pour la communication intercellulaire et d'autres fonctions cellulaires. Des r\u00e9gions c\u00e9r\u00e9brales de rat ont \u00e9t\u00e9 examin\u00e9es a la recherche de modification de phosphoty\u2010rosines prot\u00e9iques, au moyen d'une analyse de type Western blot et d'une irradiation par micro\u2010ondes afin de limiter les alt\u00e9rations post\u2010mortem, apr\u00e9s administration de 2 produits convulsivants, lithium + pilocarpine ou acide kainique. La plupart des prot\u00e9ines avec phosphotyrosines n'\u00e9taient pas modif\u00e9es par ces traitements, les auteurs ont constat\u00e9 une augmentation importante, sp\u00e9cifique de la phosphorylation de la tyrosine dans une prot\u00e9ine 40 kD. Cette \u00e9l\u00e9vation a \u00e9t\u00e9 mise en eAvidence au niveau des 3 r\u00e9gions c\u00e9r\u00e9brates examin\u00e9es, le cortex c\u00e9r\u00e9bral, \u013ehippo\u2010campe et le striatum. Cette augmentation s'est produite de fa\u00e7on brutale au moment de la mise en place d'un \u00e9tat de mal g\u00e9n\u00e9ralis\u00e9, \u013e\u00e9i\u00e9vation s'est maintenue pendant au moins 90 mn. La majorit\u00e9 de la prot\u00e9ine 40 kD avec tyrosine phosphoryi\u00e9e a \u00e9t\u00e9 trouv\u00e9e au niveau de la fraction cytosolique.
