Here it is reported that this treatment also reduces binding and phagocytosis by macrophages.<\/jats:p><jats:p><jats:bold>STUDY DESIGN AND METHODS:<\/jats:bold> Phagocytosis of mepacrine\u2010labeled PLTs by macrophages changes the latter into bright fluorescent particles easily detected by fluorescence\u2010activated cell sorting.<\/jats:p><jats:p><jats:bold>RESULTS:<\/jats:bold> In combination with conventional binding analysis it was found that binding to phorbol 12\u2010myristate 13\u2010acetate\u2013matured THP\u20101 cells is primarily regulated by PLT P\u2010selectin expression and phagocytosis by combined phosphatidylserine (PS) exposure and glycoprotein (GP) Ib\u03b1 clustering. It was found that trapping of PLT Ca<jats:sup>2+<\/jats:sup> and raising cAMP reduces phagocytosis by lowering PS exposure. Chilling of PLTs leads to an increase in binding and PS\u2010 and GPIb\u03b1\u2010mediated phagocytosis. Prior depletion of PLT energy stores prevents this increase by preserving low Ca<jats:sup>2+<\/jats:sup> concentration, PS exposure, and PS\u2010mediated phagocytosis.<\/jats:p><jats:p><jats:bold>CONCLUSION:<\/jats:bold> These data characterize the individual factors that control PLT binding and phagocytosis and might help to define conditions that improve the survival of stored PLTs after transfusion.<\/jats:p>","DOI":"10.1111\/j.1537-2995.2006.00913.x","type":"journal-article","created":{"date-parts":[[2006,7,28]],"date-time":"2006-07-28T17:38:50Z","timestamp":1154108330000},"page":"1432-1443","source":"Crossref","is-referenced-by-count":37,"title":["Platelet binding and phagocytosis by macrophages"],"prefix":"10.1111","volume":"46","author":[{"given":"Bahram A.","family":"Badlou","sequence":"first","affiliation":[]},{"given":"Ya Ping","family":"Wu","sequence":"additional","affiliation":[]},{"given":"W.
