In higher eukaryotes, genes similar to <jats:italic>SUP45<\/jats:italic> encode eukaryote release factor 1 (eRFI), which has a stop\u2010codon\u2010dependent peptidyl\u2010release activity. Using a conditional\u2010lethal mutant allele of <jats:italic>SUP45 (sup4.5\u20102)<\/jats:italic> and a combination of <jats:italic>in vivo<\/jats:italic> and <jats:italic>in vitro<\/jats:italic> approaches, we demonstrate that the product of the <jats:italic>SUP45<\/jats:italic> gene (Sup45p or eRF1) is a factor required for translation termination in yeast. A homologous <jats:italic>in vitro<\/jats:italic> assay based on suppressor\u2010tRNA\u2010mediated readthrough of stop codons is used to show that a translating lysate from a <jats:italic>sup45\u20132<\/jats:italic> mutant strain exhibits a termination defect when heated for short periods to greater than the non\u2010permissive temperature (37\u00b0C). This defect can be complemented with a purified preparation of Sup45p (eRF1) expressed in <jats:italic>Eschericha coli.<\/jats:italic> The termination defect in this strain appears to be due to an inability of the Sup45p protein to bind the ribosome, resulting <jats:italic>in vivo<\/jats:italic> in a reduced ability of Sup45p to release nascent polypeptides from the ribosome at the non\u2010permissive temperature. Cell\u2010free translation lysates from the <jats:italic>sup45\u20102<\/jats:italic> strain do not show a defect in sense\u2010codon translation at the non\u2010permissive temperature.
