For comparison the activity and kinetics of Orn\u2010T were also assayed in mouse brain homogenates. The highest activity of Orn\u2010T was found in astrocytes and in cerebral cortical neurons (5.3 \u00b1 0.5 and 5.3 \u00b1 0.4 nmol mg<jats:sup>\u20101<\/jats:sup> min<jats:sup>\u20101<\/jats:sup>, respectively) whereas the activities of Orn\u2010T in cerebellar granule cell cultures and in mouse brain were found to be about half of these values (3.1 \u00b1 0.3 and 2.8 \u00b1 0.1 nmol min<jats:sup>\u20101<\/jats:sup> mg<jats:sup>\u20101<\/jats:sup>, respectively). From a kinetic study of Orn\u2010T in the different preparations only a relatively low affinity for the enzyme with respect to ornithine was found in cerebellar granule cells, astrocytes, and whole brain [apparent K<jats:sub>m<\/jats:sub> values (at 0.5 mM \u03b3\u2010ketoglutarate): 4.7 \u00b1 0.9, 4.3 \u00b1 2.2, and 6.8 \u00b1 2.2 mM, respectively] whereas the corresponding K<jats:sub>m<\/jats:sub> value for Orn\u2010T in cerebral cortex interneurons was found to be significantly lower (apparent K<jats:sub>m<\/jats:sub>: 0.8 \u00b1 0.3 mM).
