Under normoxia (<jats:italic>P<\/jats:italic><jats:sub>O2<\/jats:sub>\u223c155 mmHg; 1 mmHg \u2248133 Pa), most NEB cells did not exhibit detectable secretory activity; however, hypoxia elicited a dose\u2010dependent (<jats:italic>P<\/jats:italic><jats:sub>O2<\/jats:sub> range 95\u201318 mmHg), tetrodotoxin (TTX)\u2010sensitive stimulation of spike\u2010like exocytotic events, indicative of vesicular amine release. High extracellular K<jats:sup>+<\/jats:sup> (50 m<jats:sc>m<\/jats:sc>) induced a secretory response similar to that elicited by severe hypoxia. Exocytosis was stimulated in normoxic NEB cells after exposure to tetraethylammonium (20 m<jats:sc>m<\/jats:sc>) or 4\u2010aminopyridine (2 m<jats:sc>m<\/jats:sc>). Hypoxia\u2010induced secretion was abolished by the non\u2010specific Ca<jats:sup>2+<\/jats:sup> channel blocker Cd<jats:sup>2+<\/jats:sup> (100 \u03bc<jats:sc>m<\/jats:sc>). Secretion was also largely inhibited by the L\u2010type Ca<jats:sup>2+<\/jats:sup> channel blocker nifedipine (2 \u03bc<jats:sc>m<\/jats:sc>), but not by the N\u2010type Ca<jats:sup>2+<\/jats:sup> channel blocker \u03c9\u2010conotoxin GVIA (1 \u03bc<jats:sc>m<\/jats:sc>). The 5\u2010HT<jats:sub>3<\/jats:sub> receptor blocker ICS 205 930 also inhibited secretion from NEB cells under hypoxia. These results suggest that hypoxia stimulates 5\u2010HT secretion from intact NEBs via inhibition of K<jats:sup>+<\/jats:sup> channels, augmentation of Na<jats:sup>+<\/jats:sup>\u2010dependent action potentials and calcium entry through L\u2010type Ca<jats:sup>2+<\/jats:sup> channels, as well as by positive feedback activation of 5\u2010HT<jats:sub>3<\/jats:sub> autoreceptors.<\/jats:p>","DOI":"10.1113\/jphysiol.2001.013071","type":"journal-article","created":{"date-parts":[[2002,7,27]],"date-time":"2002-07-27T12:45:57Z","timestamp":1027773957000},"page":"503-510","source":"Crossref","is-referenced-by-count":117,"title":["Hypoxia\u2010induced secretion of serotonin from intact pulmonary neuroepithelial bodies in neonatal rabbit"],"prefix":"10.1113","volume":"539","author":[{"given":"X.
