To obtain information about structural events that occur in axons, changes in light scattering from squid giant axons were measured during action potentials and voltage\u2010clamp steps.<\/jats:p><jats:p>2. The scattering changes were measured at several scattering angles. Because the changes in scattering divided by the resting scattering were between 10<jats:sup>\u22126<\/jats:sup> and 10<jats:sup>\u22125<\/jats:sup>, signal\u2010averaging techniques were used to increase the signal\u2010to\u2010noise ratio.<\/jats:p><jats:p>3. The scattering changes during the action potential were different at different angles. Two types were found, one at 10\u201330\u00b0 (forward angles) and the other at 60\u2013120\u00b0 (right angles).<\/jats:p><jats:p>4. At forward angles, there was a transient scattering <jats:italic>decrease<\/jats:italic> during the action potential. The time course of the change was similar to that of the action potential; this change was thought to be potential\u2010dependent.<\/jats:p><jats:p>5. At right angles, there was a transient scattering <jats:italic>increase<\/jats:italic> during the action potential followed later by a second, longer\u2010lasting increase. Indirect evidence indicated that neither component could be totally potential\u2010dependent.<\/jats:p><jats:p>6. To further analyse these effects, scattering was measured during voltage\u2010clamp steps. The changes seen during hyperpolarizing steps were presumed to be potential\u2010dependent; again two different changes were found, one at forward angles and one at right angles.<\/jats:p><jats:p>7. The potential\u2010dependent change at right angles occurred with a time course that could be approximated by a single exponential with a time constant \u03c4 = 24 \u03bcsec. The change at forward angles required two exponentials, \u03c4<jats:sub>1<\/jats:sub> = 23 \u03bcsec, \u03c4<jats:sub>2<\/jats:sub> = 900 \u03bcsec, to represent its time course.<\/jats:p><jats:p>8. The size of both potential\u2010dependent changes was proportional to the square of potential.
