{"status":"ok","message-type":"work","message-version":"1.0.0","message":{"indexed":{"date-parts":[[2026,4,1]],"date-time":"2026-04-01T19:00:50Z","timestamp":1775070050940,"version":"3.50.1"},"reference-count":40,"publisher":"American Physiological Society","issue":"5","content-domain":{"domain":[],"crossmark-restriction":false},"short-container-title":["American Journal of Physiology-Endocrinology and Metabolism"],"published-print":{"date-parts":[[2002,11,1]]},"abstract":"<jats:p> To evaluate the hypothesis that precursor supply limits gluconeogenesis (GNG) during exercise, we examined training-induced changes in glucose kinetics [rates of appearance (R<jats:sub>a<\/jats:sub>) and disappearance (R<jats:sub>d<\/jats:sub>)], oxidation (R<jats:sub>ox<\/jats:sub>), and recycling (R<jats:sub>r<\/jats:sub>) with an exogenous lactate infusion to 3.5\u20134.0 mM during rest and to pretraining 65% peak O<jats:sub>2<\/jats:sub> consumption (V\u02d9o <jats:sub>2\u2009peak<\/jats:sub>) levels during exercise. Control and clamped trials (LC) were performed at rest pre- (P<jats:sub>R<\/jats:sub>R, P<jats:sub>R<\/jats:sub>R-LC) and posttraining (P<jats:sub>O<\/jats:sub>R, P<jats:sub>O<\/jats:sub>R-LC) and during exercise pre- (P<jats:sub>R<\/jats:sub>E<jats:sub>X<\/jats:sub>) and posttraining at absolute (P<jats:sub>O<\/jats:sub>A<jats:sub>B<\/jats:sub>, P<jats:sub>O<\/jats:sub>A<jats:sub>B<\/jats:sub>-LC) and relative (P<jats:sub>O<\/jats:sub>R<jats:sub>L<\/jats:sub>, P<jats:sub>O<\/jats:sub>R<jats:sub>L<\/jats:sub>-LC) intensities. Glucose R<jats:sub>r<\/jats:sub>was not different in any rest or exercise condition. Glucose R<jats:sub>a<\/jats:sub> did not differ as a result of LC. Glucose R<jats:sub>ox<\/jats:sub> was significantly decreased with LC at P<jats:sub>O<\/jats:sub>R (0.38 \u00b1 0.03 vs. 0.56 \u00b1 0.04 mg \u00b7 kg<jats:sup>\u22121<\/jats:sup> \u00b7 min<jats:sup>\u22121<\/jats:sup>) and P<jats:sub>O<\/jats:sub>A<jats:sub>B<\/jats:sub> (3.82 \u00b1 0.51 vs. 5.0 \u00b1 0.62 mg \u00b7 kg<jats:sup>\u22121<\/jats:sup> \u00b7 min<jats:sup>\u22121<\/jats:sup>). Percent glucose R<jats:sub>d<\/jats:sub> oxidized decreased with all LC except P<jats:sub>O<\/jats:sub>R<jats:sub>L<\/jats:sub>-LC (P<jats:sub>R<\/jats:sub>R, 32%; P<jats:sub>R<\/jats:sub>R-LC, 22%; P<jats:sub>O<\/jats:sub>R, 27%; P<jats:sub>O<\/jats:sub>R-LC, 20%; P<jats:sub>O<\/jats:sub>A<jats:sub>B<\/jats:sub>, 95%; P<jats:sub>O<\/jats:sub>A<jats:sub>B<\/jats:sub>-LC, 77%), which resulted in a significant increase in oxidation from alternative carbohydrate (CHO) sources at rest and P<jats:sub>O<\/jats:sub>A<jats:sub>B<\/jats:sub>. We conclude that 1) increased arterial [lactate] did not increase glucose R<jats:sub>r<\/jats:sub>measured during rest or exercise after training, 2) glucose disposal or production did not change with increased precursor supply, and 3) infusion of exogenous CHO in the form of lactate resulted in the decrease of glucose R<jats:sub>ox<\/jats:sub>.
