Studies revealed an important role of microRNAs (miRNAs) in multiple cancers, including breast cancer. In the present study, we evaluated the role and function of miR-641 in breast cancer. Methods. The expression level of miR-641 in breast cancer cell lines (Hs-578T, MCF7, HCC1937, and MAD-MB-231) was determined by real-time PCR. Functional analyses, including CCK-8 assay, transwell assay, wound-healing assay, and apoptosis detection, were carried out to explore the roles of miRNA-641 in malignant behaviors of breast cancer. Luciferase report assay was used to investigate the regulatory association of miRNA-641 with its potential targets. Results. The expression levels of miR-641 were downregulated, while the expression levels of nuclear casein kinase and cyclin-dependent kinase substrate 1 (NUCKS1) were increased in breast cancer cell lines. The in vitro results showed that miR-641 repressed proliferation and migration\/invasion and promoted apoptosis of breast cancer cells. NUCKS1, a positive regulator of phosphatidylinositol-3-kinases (PI3K)\/protein-serine-threonine kinase (AKT) pathway, was confirmed as a direct target of miR-641. The of treatment of the PI3K agonist, 740Y-P, could abrogate the antioncogenic potentials of miR-641 in breast cancer cells. Conclusion. miR-641 functioned as a tumor suppressor through the PI3K\/AKT signaling pathway via targeting NUCKS1 in breast cancer.<\/jats:p>","DOI":"10.1155\/2022\/5203839","type":"journal-article","created":{"date-parts":[[2022,1,12]],"date-time":"2022-01-12T19:35:18Z","timestamp":1642016118000},"page":"1-18","source":"Crossref","is-referenced-by-count":12,"title":["miR-641 Inhibited Cell Proliferation and Induced Apoptosis by Targeting NUCKS1\/PI3K\/AKT Signaling Pathway in Breast Cancer"],"prefix":"10.1155","volume":"2022","author":[{"ORCID":"https:\/\/orcid.org\/0000-0003-4834-8447","authenticated-orcid":true,"given":"Li","family":"Li","sequence":"first","affiliation":[{"name":"Department of General Surgery, Nanjing Medical University Affiliated Cancer Hospital Cancer Institute of Jiangsu Province, Baiziting 42, Nanjing 210009, China"},{"name":"The Fourth Clinical School of Nanjing Medical University, Nanjing 210009, China"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"ORCID":"https:\/\/orcid.org\/0000-0001-5710-2233","authenticated-orcid":true,"given":"Da","family":"Wei","sequence":"additional","affiliation":[{"name":"Department of General Surgery, Nanjing Medical University Affiliated Cancer Hospital Cancer Institute of Jiangsu Province, Baiziting 42, Nanjing 210009, China"},{"name":"The Fourth Clinical School of Nanjing Medical University, Nanjing 210009, China"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"ORCID":"https:\/\/orcid.org\/0000-0002-1554-0339","authenticated-orcid":true,"given":"Junying","family":"Zhang","sequence":"additional","affiliation":[{"name":"Department of General Surgery, Nanjing Medical University Affiliated Cancer Hospital Cancer Institute of Jiangsu Province, Baiziting 42, Nanjing 210009, China"},{"name":"Center of Clinical Laboratory, Cancer Institute of Jiangsu Province, Nanjing Medical University Affiliated Cancer Hospital, Nanjing, China"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"ORCID":"https:\/\/orcid.org\/0000-0002-3431-5030","authenticated-orcid":true,"given":"Rong","family":"Deng","sequence":"additional","affiliation":[{"name":"Department of General Surgery, Nanjing Medical University Affiliated Cancer Hospital Cancer Institute of Jiangsu Province, Baiziting 42, Nanjing 210009, China"},{"name":"The Fourth Clinical School of Nanjing Medical University, Nanjing 210009, China"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"ORCID":"https:\/\/orcid.org\/0000-0003-2016-4216","authenticated-orcid":true,"given":"Jinhai","family":"Tang","sequence":"additional","affiliation":[{"name":"Department of General Surgery, Nanjing Medical University Affiliated Cancer Hospital Cancer Institute of Jiangsu Province, Baiziting 42, Nanjing 210009, China"},{"name":"Department of General Surgery, The First Affiliated Hospital with Nanjing Medical University, Nanjing, Jiangsu, China"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"ORCID":"https:\/\/orcid.org\/0000-0003-4503-4709","authenticated-orcid":true,"given":"Dongming","family":"Su","sequence":"additional","affiliation":[{"name":"Center of Metabolic Research, Nanjing Medical University, Nanjing, China"}],"role":[{"role":"author","vocabulary":"crossref"}]}],"member":"311","reference":[{"key":"1","doi-asserted-by":"publisher","DOI":"10.1186\/s12940-020-00670-2"},{"key":"2","doi-asserted-by":"publisher","DOI":"10.1016\/j.breast.2011.04.007"},{"key":"3","doi-asserted-by":"publisher","DOI":"10.1007\/s12282-018-0848-6"},{"key":"4","doi-asserted-by":"publisher","DOI":"10.12688\/f1000research.18888.1"},{"key":"5","doi-asserted-by":"publisher","DOI":"10.1007\/s12032-017-1071-6"},{"key":"6","doi-asserted-by":"publisher","DOI":"10.1186\/s13045-019-0798-2"},{"key":"7","doi-asserted-by":"publisher","DOI":"10.21037\/cco.2018.05.05"},{"key":"8","doi-asserted-by":"publisher","DOI":"10.1371\/journal.pone.0173721"},{"key":"9","doi-asserted-by":"publisher","DOI":"10.1186\/s12967-015-0425-0"},{"key":"10","doi-asserted-by":"publisher","DOI":"10.3747\/co.25.3752"},{"key":"11","doi-asserted-by":"publisher","DOI":"10.1007\/s10555-016-9637-x"},{"key":"12","doi-asserted-by":"publisher","DOI":"10.1016\/j.joim.2019.02.001"},{"key":"13","doi-asserted-by":"publisher","DOI":"10.1016\/j.joim.2019.03.003"},{"key":"14","doi-asserted-by":"publisher","DOI":"10.1158\/1078-0432.CCR-16-3001"},{"key":"15","doi-asserted-by":"crossref","first-page":"264","DOI":"10.2174\/0929867321666141106124315","article-title":"Targeting the NF\u03baB signaling pathways for breast cancer prevention and therapy","volume":"22","author":"W.
