It has long been used in folk medicine as a remedy for various ailments. This is the first report to demonstrate the protective antioxidant and anti-inflammatory properties of aqueous methanolic extracts derived from the aerial parts (MtAPV) and roots (MtRV) of<jats:italic>in vitro<\/jats:italic>grown plants on human umbilical vein endothelial cells (HUVECs). It describes the influence of the tested extracts on the expression of antioxidant (HO-1, NQO1,<jats:italic>NRF2<\/jats:italic>,<jats:italic>kEAP1<\/jats:italic>, and<jats:italic>GCLC<\/jats:italic>) and inflammation-related genes (<jats:italic>IL-1\u03b1<\/jats:italic>,<jats:italic>IL-1\u03b2<\/jats:italic>,<jats:italic>IL-6<\/jats:italic>,<jats:italic>TNF-\u03b1<\/jats:italic>, and<jats:italic>IFN-\u03b3<\/jats:italic>) in cells stimulated with H<jats:sub>2<\/jats:sub>O<jats:sub>2<\/jats:sub>or LPS, respectively. In addition,<jats:italic>M. trifoliata<\/jats:italic>extracts were found to moderately affect the growth of certain bacterial and fungal pathogens, with the strongest antibacterial effect found against<jats:italic>Pseudomonas aeruginosa<\/jats:italic>and<jats:italic>Enterococcus faecalis<\/jats:italic>.<jats:italic>M. trifoliata<\/jats:italic>extracts demonstrated protective effects against mitochondrial DNA (mtDNA) and nuclear DNA (nDNA) damage caused by ROS, decreasing the numbers of mtDNA lesions in the<jats:italic>ND1<\/jats:italic>and<jats:italic>ND2<\/jats:italic>genes and nDNA damage in the<jats:italic>TP53<\/jats:italic>and<jats:italic>HPRT1<\/jats:italic>genes and reducing cleavage in PARP1- and<jats:italic>\u03b3<\/jats:italic>-H2A.X-positive cells. The root extract of<jats:italic>in vitro M. trifoliata<\/jats:italic>(MtRV) appears to have better anti-inflammatory, antioxidant, antimicrobial, and protective properties than the extract from the aerial part (MtAPV).
