{"status":"ok","message-type":"work","message-version":"1.0.0","message":{"indexed":{"date-parts":[[2025,3,6]],"date-time":"2025-03-06T22:51:19Z","timestamp":1741301479790,"version":"3.38.0"},"reference-count":0,"publisher":"Walter de Gruyter GmbH","issue":"1","content-domain":{"domain":[],"crossmark-restriction":false},"short-container-title":["Nematol"],"published-print":{"date-parts":[[2006]]},"abstract":"<jats:sec>\n\t\t\t\t\t<jats:title>Abstract<\/jats:title>\n\t\t\t\t\t<jats:p>Capillary gel electrophoresis was used to characterise 49 Portuguese isolates\n\t\t\t\t\t\tof potato cyst nematodes, three of Globodera pallida and 46 of G.\n\t\t\t\t\t\trostochiensis, and eight isolates representative of pathotypes defined for\n\t\t\t\t\t\tthese two species (Ro1, Ro2, Ro3, Ro4 and Ro5 for G. rostochiensis and Pa1,\n\t\t\t\t\t\tPa2 and Pa3 for G. pallida). Protein extracts of four samples of 50 cysts\n\t\t\t\t\t\tper isolate were separated using a Beckman eCAP SDS 14-200 kit, in a\n\t\t\t\t\t\tcapillary of 57 cm length and 100 \u03bc m internal diameter and with a run time\n\t\t\t\t\t\tof 40 min. Reproducible protein profiles were obtained and the isolates were\n\t\t\t\t\t\tcompared taking into account the relative migration time and area of each\n\t\t\t\t\t\tpeak recorded by the Gold Software Data System (Beckman). Euclidian\n\t\t\t\t\t\tdistances between isolates were calculated using peak area data and a\n\t\t\t\t\t\tdendrogram was constructed according to the UPGMA method. Portuguese\n\t\t\t\t\t\tisolates showed great variability and it seems that protein composition is\n\t\t\t\t\t\tnot related to their geographic origin. Protein profiles of each pathotype\n\t\t\t\t\t\twere obtained and some of the proteins seem to be specific for each\n\t\t\t\t\t\tpathotype.
