Hence, the infectious capacity of the pathogen is closely related to its ability to interact with host proteins. Here, we show that hubs in the host-pathogen interactome are isolated in the pathogen network by adapting the geometry of the interacting interfaces. An imperfect mimicry of the eukaryotic interfaces allows pathogen proteins to actively bind to the host\u2019s target while preventing deleterious effects on the pathogen interactome. Understanding how bacteria recognize eukaryotic proteins may pave the way for the rational design of new antibiotic molecules.<\/jats:p>","DOI":"10.1371\/journal.pcbi.1008395","type":"journal-article","created":{"date-parts":[[2020,12,4]],"date-time":"2020-12-04T15:12:28Z","timestamp":1607094748000},"page":"e1008395","update-policy":"https:\/\/doi.org\/10.1371\/journal.pcbi.corrections_policy","source":"Crossref","is-referenced-by-count":12,"title":["Bacteria use structural imperfect mimicry to hijack the host interactome"],"prefix":"10.1371","volume":"16","author":[{"given":"Natalia Sanchez","family":"de Groot","sequence":"first","affiliation":[]},{"ORCID":"https:\/\/orcid.org\/0000-0001-6567-3474","authenticated-orcid":true,"given":"Marc","family":"Torrent Burgas","sequence":"additional","affiliation":[]}],"member":"340","published-online":{"date-parts":[[2020,12,4]]},"reference":[{"key":"pcbi.1008395.ref001","doi-asserted-by":"crossref","first-page":"16022","DOI":"10.1038\/nmicrobiol.2016.22","article-title":"Self-organized patchiness facilitates survival in a cooperatively growing Bacillus subtilis population.","volume":"1","author":"C Ratzke","year":"2016","journal-title":"Nat Microbiol"},{"issue":"1","key":"pcbi.1008395.ref002","doi-asserted-by":"crossref","first-page":"63","DOI":"10.1016\/j.chom.2013.05.005","article-title":"Pathogens use structural mimicry of native host ligands as a mechanism for host receptor engagement","volume":"14","author":"N Drayman","year":"2013","journal-title":"Cell Host Microbe"},{"issue":"11","key":"pcbi.1008395.ref003","doi-asserted-by":"crossref","first-page":"4413","DOI":"10.1128\/IAI.05622-11","article-title":"Insight into bacterial virulence mechanisms against host immune response via the Yersinia pestis-human protein-protein interaction network","volume":"79","author":"H Yang","year":"2011","journal-title":"Infect Immun"},{"issue":"11","key":"pcbi.1008395.ref004","doi-asserted-by":"crossref","first-page":"3036","DOI":"10.1074\/mcp.M113.029041","article-title":"Novel Burkholderia mallei virulence factors linked to specific host-pathogen protein interactions","volume":"12","author":"V Memisevic","year":"2013","journal-title":"Mol Cell Proteomics"},{"issue":"6","key":"pcbi.1008395.ref005","doi-asserted-by":"crossref","first-page":"343","DOI":"10.1038\/nrmicro3456","article-title":"Secretion systems in Gram-negative bacteria: structural and mechanistic insights","volume":"13","author":"TR Costa","year":"2015","journal-title":"Nat Rev Microbiol"},{"issue":"7","key":"pcbi.1008395.ref006","doi-asserted-by":"crossref","first-page":"425","DOI":"10.1111\/tra.12488","article-title":"The outer membrane vesicles: Secretion system type zero","volume":"18","author":"A Guerrero-Mandujano","year":"2017","journal-title":"Traffic"},{"issue":"24","key":"pcbi.1008395.ref007","doi-asserted-by":"crossref","first-page":"8710","DOI":"10.1073\/pnas.0500879102","article-title":"Competence-programmed predation of noncompetent cells in the human pathogen Streptococcus pneumoniae: genetic requirements","volume":"102","author":"S Guiral","year":"2005","journal-title":"Proc Natl Acad Sci U S A"},{"key":"pcbi.1008395.ref008","doi-asserted-by":"crossref","first-page":"14092","DOI":"10.1038\/ncomms14092","article-title":"Centrality in the host-pathogen interactome is associated with pathogen fitness during infection.","volume":"8","author":"N Crua Asensio","year":"2017","journal-title":"Nat Commun."},{"issue":"1","key":"pcbi.1008395.ref009","doi-asserted-by":"crossref","first-page":"2312","DOI":"10.1038\/s41467-018-04632-8","article-title":"Network biology discovers pathogen contact points in host protein-protein interactomes","volume":"9","author":"H Ahmed","year":"2018","journal-title":"Nat Commun"},{"issue":"6833","key":"pcbi.1008395.ref010","doi-asserted-by":"crossref","first-page":"41","DOI":"10.1038\/35075138","article-title":"Lethality and centrality in protein networks","volume":"411","author":"H Jeong","year":"2001","journal-title":"Nature"},{"issue":"1","key":"pcbi.1008395.ref011","doi-asserted-by":"crossref","first-page":"73","DOI":"10.1007\/s11693-013-9123-5","article-title":"The organisational structure of protein networks: revisiting the centrality-lethality hypothesis","volume":"8","author":"K Raman","year":"2014","journal-title":"Syst Synth Biol"},{"issue":"6379","key":"pcbi.1008395.ref012","doi-asserted-by":"crossref","first-page":"543","DOI":"10.1038\/357543a0","article-title":"Proteins.
