The Cabo Verde archipelago harbors a rich native plant diversity, most of which are used with medicinal purposes. <jats:bold>Methods<\/jats:bold>The present study investigated the <jats:italic>in vitro<\/jats:italic> antiplasmodial activities of four native plants from Cabo Verde. Aqueous and ethanolic extracts of these plants were tested <jats:italic>in vitro<\/jats:italic> against both chloroquine-sensitive (3D7) and chloroquine-resistant (Dd2) <jats:italic>P. falciparum<\/jats:italic> strains using a SYBR Green detection method, and their cytotoxicity was evaluated in Caco and PLP2 cells by using a sulforhodamine B colorimetric assay. <jats:bold>Results<\/jats:bold>An ethanolic extract of <jats:italic>Artemisia<\/jats:italic> <jats:italic>gorgonum<\/jats:italic> and an aqueous extract of <jats:italic>Tamarix senegalensis<\/jats:italic> exhibited high antiplasmodial activity (EC<jats:sub>50 <\/jats:sub>&lt; 5 \u03bcg\/mL) without cytotoxicity. Extracts of <jats:italic>Lavandula rotundifolia <\/jats:italic>and <jats:italic>Sideroxylon marginatum<\/jats:italic> exhibited moderate activity, with EC<jats:sub>50<\/jats:sub> values ranging from 10-30 \u03bcg\/mL.
