{"status":"ok","message-type":"work","message-version":"1.0.0","message":{"institution":[{"name":"Authorea, Inc."}],"indexed":{"date-parts":[[2025,6,3]],"date-time":"2025-06-03T16:35:52Z","timestamp":1748968552092,"version":"3.41.0"},"posted":{"date-parts":[[2021,4,8]]},"group-title":"Preprints","reference-count":0,"publisher":"Wiley","content-domain":{"domain":[],"crossmark-restriction":false},"short-container-title":[],"accepted":{"date-parts":[[2021,4,8]]},"abstract":"<jats:p id=\"p1\">Background and purpose Vasoactive intestinal peptide (VIP), acting on\nboth VPAC and VPAC receptors, is a key\nmodulator of hippocampal synaptic transmission, pyramidal cell\nexcitability and synaptic plasticity phenomena, like long-term\ndepression (LTD), partly through modulation GABAergic disinhibitory\ncircuits. VIP effects on LTP and the involvement of disinhibition were\nscarcely investigated. Experimental approach The influence of endogenous\nVIP on CA1 LTP induced by TBS was evaluated in the CA1 area of\nhippocampal slices using field-excitatory electrophysiological\nrecordings from young-adult Wistar rats using selective\nVPAC and VPAC antagonists.\nPhosphorylation of GluA1 AMPA receptor subunits and Kv4.2 potassium\nchannels was evaluated in hippocampal membranes obtained from such\nslices by Western blot. Key results Here we show that VIP, acting on\nVPAC (but not VPAC) receptors, is an\nendogenous inhibitor of hippocampal LTP induced by theta-burst\nstimulation (TBS) in the CA1 area of the hippocampus of young adult\nWistar rats. This effect is dependent on GABAergic transmission and\nrelies on the integrity of NMDA and CaMKII-dependent LTP expression\nmechanisms but not on PKA and PKC activity. Furthermore, it regulates\nthe expression and Serphosphorylation of Kv4.2\npotassium channels responsible for the A-current while inhibiting\nphosphorylation of Kv4.2 on Thr. Conclusions and\nimplications Altogether this suggests that endogenous VIP controls the\nexpression of hippocampal CA1 LTP by regulating disinhibition through\nactivation of VPAC receptors in interneurons.
