{"status":"ok","message-type":"work","message-version":"1.0.0","message":{"indexed":{"date-parts":[[2026,5,18]],"date-time":"2026-05-18T11:25:51Z","timestamp":1779103551538,"version":"3.51.4"},"posted":{"date-parts":[[2022,10,26]]},"reference-count":0,"publisher":"Wiley","content-domain":{"domain":[],"crossmark-restriction":false},"short-container-title":[],"abstract":"<jats:p>Background and purpose: Montelukast, an antagonist of the cysteinyl\nleukotrienes receptor 1, has been proposed for repurposing for the\ntreatment of neurodegenerative disorders, including Alzheimer\u2019s disease.\nClinical trials are ongoing but the mechanisms supporting this\nrepurposing are still poorly understood. Experimental Approach: Taking\nadvantage of proteomics datasets deposited in public repositories, data\nfrom mouse brain and a neuronal chicken model exposed to the drug were\nreinterpreted in view of the repurposing proposal. Key Results:\nMontelukast increases the levels of presenilin 1, nicastrin, neprilysin,\nand insulin-degrading enzyme, all of which are involved in the amyloid\naggregation and clearance processes. Hexokinase 1, malate and isocitrate\ndehydrogenate enzymes, from central metabolism pathways, are also\naffected. Conclusions and Implications: The data suggest that\nmontelukast is a modulator of the amyloid clearance process, favouring\nthe removal of aggregates and counterbalancing the overall amyloidogenic\nprocess. Montelukast also acts on energy supply pathways, compensating\nthe ageing-associated decrease of the basal cell metabolism.
