In life science, such sources of error might be the broad variety of additives such as dithiothreitol (DTT) used to preserve enzyme stability during in vitro reactions. Using two different assays that can sense strand interruptions in double stranded DNA, we here show that DTT is able to introduce nicks in the DNA backbone. DTT was furthermore shown to facilitate the immobilization of fluorescent DNA on an NHS-ester functionalized glass surface. Such reactions may in particular impact the readout from single molecule detection studies and other ultrasensitive assays. This was highlighted by the finding that DTT markedly decreased the signal to noise ratio in a DNA sensor based assay with single molecule resolution.<\/jats:p>","DOI":"10.3390\/s17061201","type":"journal-article","created":{"date-parts":[[2017,5,24]],"date-time":"2017-05-24T07:29:42Z","timestamp":1495610982000},"page":"1201","update-policy":"https:\/\/doi.org\/10.3390\/mdpi_crossmark_policy","source":"Crossref","is-referenced-by-count":24,"title":["The Effects of Dithiothreitol on DNA"],"prefix":"10.3390","volume":"17","author":[{"given":"S\u00f8ren","family":"Fjelstrup","sequence":"first","affiliation":[{"name":"Department of Molecular Biology and Genetics, Aarhus University, 8000 Aarhus, Denmark"},{"name":"Interdisciplinary Nanoscience Center (iNANO), Aarhus University, 8000 Aarhus, Denmark"}],"role":[{"vocabulary":"crossref","role":"author"}]},{"given":"Marie","family":"Andersen","sequence":"additional","affiliation":[{"name":"Department of Molecular Biology and Genetics, Aarhus University, 8000 Aarhus, Denmark"}],"role":[{"vocabulary":"crossref","role":"author"}]},{"given":"Jonas","family":"Thomsen","sequence":"additional","affiliation":[{"name":"Department of Molecular Biology and Genetics, Aarhus University, 8000 Aarhus, Denmark"}],"role":[{"vocabulary":"crossref","role":"author"}]},{"given":"Jing","family":"Wang","sequence":"additional","affiliation":[{"name":"Department of Molecular Biology and Genetics, Aarhus University, 8000 Aarhus, Denmark"}],"role":[{"vocabulary":"crossref","role":"author"}]},{"given":"Magnus","family":"Stougaard","sequence":"additional","affiliation":[{"name":"Department of Pathology, Aarhus University Hospital, 8000 Aarhus, Denmark"}],"role":[{"vocabulary":"crossref","role":"author"}]},{"given":"Finn","family":"Pedersen","sequence":"additional","affiliation":[{"name":"Department of Molecular Biology and Genetics, Aarhus University, 8000 Aarhus, Denmark"},{"name":"Interdisciplinary Nanoscience Center (iNANO), Aarhus University, 8000 Aarhus, Denmark"}],"role":[{"vocabulary":"crossref","role":"author"}]},{"ORCID":"https:\/\/orcid.org\/0000-0002-2052-9724","authenticated-orcid":false,"given":"Yi-Ping","family":"Ho","sequence":"additional","affiliation":[{"name":"Department of Molecular Biology and Genetics, Aarhus University, 8000 Aarhus, Denmark"},{"name":"Interdisciplinary Nanoscience Center (iNANO), Aarhus University, 8000 Aarhus, Denmark"},{"name":"Division of Biomedical Engineering, Department of Electronic Engineering, The Chinese University of Hong Kong, Shatin, NT, Hong Kong SAR, China"}],"role":[{"vocabulary":"crossref","role":"author"}]},{"given":"Marianne","family":"Hede","sequence":"additional","affiliation":[{"name":"Zymonostics ApS, 8000 Aarhus, Denmark"}],"role":[{"vocabulary":"crossref","role":"author"}]},{"given":"Birgitta","family":"Knudsen","sequence":"additional","affiliation":[{"name":"Department of Molecular Biology and Genetics, Aarhus University, 8000 Aarhus, Denmark"},{"name":"Interdisciplinary Nanoscience Center (iNANO), Aarhus University, 8000 Aarhus, Denmark"}],"role":[{"vocabulary":"crossref","role":"author"}]}],"member":"1968","published-online":{"date-parts":[[2017,5,24]]},"reference":[{"key":"ref_1","doi-asserted-by":"crossref","first-page":"223","DOI":"10.1021\/nn800509b","article-title":"Single-molecule detection of human topoisomerase I cleavage-ligation activity","volume":"3","author":"Stougaard","year":"2009","journal-title":"ACS Nano"},{"key":"ref_2","doi-asserted-by":"crossref","first-page":"461","DOI":"10.1038\/nmeth.1459","article-title":"Direct detection of DNA methylation during single-molecule, real-time sequencing","volume":"7","author":"Flusberg","year":"2010","journal-title":"Nat.
