Because of its small size, low cost, and being easy to hold and operate, it can be used as an alternative tool for large microscopes in resource-poor or remote areas, which is of great significance for the diagnosis, treatment, and prevention of diseases. To improve the low-resolution characteristics of the existing lensless shadow imaging systems and to meet the high-resolution needs of point-of-care testing, here, we propose a high-precision on-chip microscope based on in-line holographic technology. We demonstrated the ability of the iterative phase recovery algorithm to recover sample information and evaluated it with image quality evaluation algorithms with or without reference. The results showed that the resolution of the holographic image after iterative phase recovery is 1.41 times that of traditional shadow imaging. Moreover, we used machine learning tools to identify and count the mixed samples of mouse ascites tumor cells and micro-particles that were iterative phase recovered. The results showed that the on-chip cell counter had high-precision counting characteristics as compared with manual counting of the microscope reference image. Therefore, the proposed high-precision lensless microscope on a chip based on in-line holographic imaging provides one promising solution for future point-of-care testing (POCT).<\/jats:p>","DOI":"10.3390\/s21030720","type":"journal-article","created":{"date-parts":[[2021,1,21]],"date-time":"2021-01-21T11:49:40Z","timestamp":1611229780000},"page":"720","update-policy":"https:\/\/doi.org\/10.3390\/mdpi_crossmark_policy","source":"Crossref","is-referenced-by-count":18,"title":["High-Precision Lensless Microscope on a Chip Based on In-Line Holographic Imaging"],"prefix":"10.3390","volume":"21","author":[{"ORCID":"https:\/\/orcid.org\/0000-0002-2364-0479","authenticated-orcid":false,"given":"Xiwei","family":"Huang","sequence":"first","affiliation":[{"name":"Ministry of Education Key Lab of RF Circuits and Systems, Hangzhou Dianzi University, Hangzhou 310018, China"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"ORCID":"https:\/\/orcid.org\/0000-0003-0806-5872","authenticated-orcid":false,"given":"Yangbo","family":"Li","sequence":"additional","affiliation":[{"name":"Ministry of Education Key Lab of RF Circuits and Systems, Hangzhou Dianzi University, Hangzhou 310018, China"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"given":"Xuefeng","family":"Xu","sequence":"additional","affiliation":[{"name":"Ministry of Education Key Lab of RF Circuits and Systems, Hangzhou Dianzi University, Hangzhou 310018, China"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"given":"Renjie","family":"Wang","sequence":"additional","affiliation":[{"name":"Ministry of Education Key Lab of RF Circuits and Systems, Hangzhou Dianzi University, Hangzhou 310018, China"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"given":"Jiangfan","family":"Yao","sequence":"additional","affiliation":[{"name":"Ministry of Education Key Lab of RF Circuits and Systems, Hangzhou Dianzi University, Hangzhou 310018, China"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"given":"Wentao","family":"Han","sequence":"additional","affiliation":[{"name":"Ministry of Education Key Lab of RF Circuits and Systems, Hangzhou Dianzi University, Hangzhou 310018, China"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"given":"Maoyu","family":"Wei","sequence":"additional","affiliation":[{"name":"Ministry of Education Key Lab of RF Circuits and Systems, Hangzhou Dianzi University, Hangzhou 310018, China"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"given":"Jin","family":"Chen","sequence":"additional","affiliation":[{"name":"Ministry of Education Key Lab of RF Circuits and Systems, Hangzhou Dianzi University, Hangzhou 310018, China"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"given":"Weipeng","family":"Xuan","sequence":"additional","affiliation":[{"name":"Ministry of Education Key Lab of RF Circuits and Systems, Hangzhou Dianzi University, Hangzhou 310018, China"}],"role":[{"role":"author","vocabulary":"crossref"}]},{"given":"Lingling","family":"Sun","sequence":"additional","affiliation":[{"name":"Ministry of Education Key Lab of RF Circuits and Systems, Hangzhou Dianzi University, Hangzhou 310018, China"}],"role":[{"role":"author","vocabulary":"crossref"}]}],"member":"1968","published-online":{"date-parts":[[2021,1,21]]},"reference":[{"key":"ref_1","doi-asserted-by":"crossref","first-page":"1016","DOI":"10.1364\/OE.19.001016","article-title":"Spatial light interference microscopy (SLIM)","volume":"19","author":"Wang","year":"2011","journal-title":"Opt.
